I think it started with their observed effects on apo-AI and beta amyloid. But since has expanded to other mechanisms as well. The RVX posters/publication page has a bunch of resources documenting the newer stuff on inflammation, etc. apo-AI and HDL have long been thought to play a role. Alkaline phosphatase may play a role too. Effect on amyloid beta 40 summarized in this post . Original AB40 results were from ASSERT long ago.
Apabetalone decreases plasma ALP, increases plasma amyloid beta 40, decreases inflammation and increases plasma apoAI/HDL. All consistent with apabetalone being preventative for Alzheimer's disease and dementia.
